Mitochondria, tiny structures within each cell that regulate metabolism and energy use, may be a promising new target for cancer therapy, according to a new study. Manipulation of two biochemical signals that regulate the numbers of mitochondria in cells could shrink human lung cancers transplanted into mice, a team of Chicago researchers report in the journal FASEB.

Within each cell, mitochondria are constantly splitting in two, a process called fission, and merging back into one, called fusion. Before a cell can divide, the mitochondria must increase their numbers through fission and separate into two piles, one for each cell.

By reversing an imbalance of the signals that regulate fusion and fission in rapidly dividing cancer cells, researchers were able to dramatically reduce cell division, thus preventing the rapid cell proliferation that is a hallmark of cancer growth. Increasing production of the signal that promotes mitochondrial fusion caused tumors to shrink to one-third of their original size. Treatment with a molecule that inhibits fission reduced tumor size by more than half.

"We found that human lung cancer cell lines have an imbalance of signals that tilts them towards mitochondrial fission," said Stephen L. Archer, MD, the Harold Hines Jr. Professor of Medicine at the University of Chicago Medicine and senior author of the study. "By boosting the fusion signal or blocking the fission signal we were able to tip the balance the other way, reducing cancer cell growth and increasing cell death. We believe this provides a promising new approach to cancer treatment."

"This could be a potential new Achilles' heel for cancer cells," said the study's lead author, Jalees Rehman, MD, an associate professor of medicine and pharmacology at the University of Illinois at Chicago. "Many anticancer drugs target cell division. Our work shifts the focus to a distinct but necessary step: mitochondrial division. The cell division cycle comes to a halt if the mitochondria are prevented from dividing. This new therapy may be especially useful in cancers which become resistant to conventional chemotherapy that directly targets the cycle."

The researchers found that the mitochondrial networks within several different lung cancer cell lines were highly fragmented, compared to normal lung cells. Cancer cells had low levels of mitofusin-2 (Mfn-2), a protein that promotes fusion by tethering adjacent mitochondria, and high levels of dynamin-related protein (Drp-1), which initiates fission by encircling the organelle and squeezing it into two discrete fragments. The Drp-1 in cancer cells also tended to be in its most active form.

The researchers tested several ways to enhance fusion and restore the mitochondrial network, both in cell culture and in animal models. They used gene therapy to increase the expression of Mfn-2, injected a small molecule (mdivi-1) that inhibits Drp-1, and used genetic techniques to block the production of Drp-1. All three interventions markedly reduced mitochondrial fragmentation, increased networking and reduced cancer cell growth.

Although the authors identify mitochondrial fission and Drp-1 activation as a potential therapeutic target in lung cancer, "this is not a cure," Archer emphasized. The treatment drastically reduced tumor size but the tumors did not completely disappear. They continued to use high levels of glucose as fuel, a hallmark of cancer metabolism that can be seen on PET scans. "This remnant could be either a central cluster of cancer stem cells," Archer said, "or an inflammatory response, the immune system infiltrating the tumor."

"Inhibiting mitochondrial fission", Archer said, "did not show any significant toxicity in mice or rats, so we are quite optimistic that our findings can lead to the development of novel, clinically feasible therapies."

The substances used to block fusion are commercially available for research purposes, but they have not been tested in humans. Mdivi-1 has been used in animals to prevent kidney injury.

Although the focus on mitochondria is fairly new to cancer biologistsdespite a flurry of interest in the 1920s stimulated by the German Nobel Prize laureate Otto Warburgthis organelle has long been a central focus for physicians and scientists interested in muscle biology, especially cardiac muscle.

Archer, a cardiologist, specializes in pulmonary hypertension. In this disorder, as in cancer, excessive cellular growth causes disease. The death of his cousin and close friend from lung cancer made him start thinking about the connections. Rehman is a German scientist and became interested in studying mitochondria after reading some of the historical Warburg papers in German.

The fact that two cardiologists, Archer and Rehman, decided to study cancer and collaborated with a team of basic scientists, a cancer physician and a pathologist is "an indicator of how interconnected modern biomedical research has become," Rehman said.

UK man appeals Twitter 'joke' conviction

A British man who was convicted and fined for jokingly posting a message on Twitter saying he would blow up an airport is appealing his conviction.

Paul Chambers was fined 385 pounds ($612) and ordered to pay legal costs after he posted a message on his Twitter feed saying he would blow up Robin Hood airport in northern England if his flight was delayed.

Chambers argued the tweet sent in Jan. 2010 was obviously a joke aimed at his Twitter followers but the courts decided that the message was menacing and convicted him under legislation that makes it illegal to send menacing or offensive messages electronically.

The case will be heard Wednesday at London's High Court.

Dr. Oz's 'Transformation Nation' Health Effort Nets 1 Million Participants

NEW YORK -- Television already has "The Biggest Loser." Dr. Mehmet Oz is looking for the biggest number of losers.

"The Dr. Oz Show" said Monday that it had netted its 1 millionth participant in its "transformation nation" health effort, and the number is climbing. One of those people will win a $1 million prize in May.

Since September, Oz has urged viewers to participate in his health challenge, done together with Weight Watchers. The number of registrants has increased steadily to a point that Oz said he'd never imagined the program would reach.

"It is one of the most rewarding experiences of my life," Oz said.

Oz's program has seven steps, starting with the simplest: Tell a friend to get some moral support. Participants are asked to register with Weight Watchers and go to a center to have their body mass index calculated. Other steps include connecting with a doctor, learning your family's health history, getting more sleep, managing stress better and starting new fitness habits.

A sample of 96,000 registrants who had weighed in at Weight Watchers determined they had collectively lost 672,000 pounds, or an average of 7 pounds per person, the show said.

The show will select 10 finalists that it believes best embody the effort ? not necessarily those who lose the most weight ? and viewers will choose a favorite this spring for the $1 million prize. People need to register by Feb. 26 to be eligible.

Participants are sent weekly email reminders by the show of things they need to do to reach their goals, tailored to their individual circumstances.

"Fundamentally, it's about making it easier to do the right thing," Oz said.

Oz also said he hoped the project would be a model for using TV to accomplish worthwhile goals.

(AP) ? The Obama administration on Saturday called for stepped up U.S.-European cooperation to isolate tyrannies like the Assad regime in Syria, promote democracy in the Arab World and beyond and repair damage from the global financial crisis.

And, as America shifts its primary strategic focus to Asia and reduces its military presence in Europe, President Barack Obama's top two national security aides ? attending an international security conference here together in a demonstration of Washington's resolve ? reassured the continent that it remains deeply relevant to U.S. interests as well as its partner of "first resort" in dealing with global hotspots.

U.S. Secretary of State Hillary Rodham Clinton said Americans and Europeans must send a clear and common message to tyrants that they must respect the rights of their people. She spoke as violence flared anew in Syria ahead of an expected vote on a U.N. Security Council resolution on the matter later Saturday.

"As a tyrant in Damascus brutalizes his own people, America and Europe stand shoulder to shoulder," she said, referring to Syrian President Bashar Assad.

"We are united, alongside the Arab League, in demanding an end to the bloodshed and a democratic future for Syria. And we are hopeful that at 10 a.m. Eastern Standard Time in New York the Security Council will express the will of the international community," she said.

President Barack Obama, in a strongly worded statement Saturday, condemned Assad's "unspeakable assault" and called for his ouster.

"Assad must halt his campaign of killing and crimes against his own people now. He must step aside and allow a democratic transition to proceed immediately," the president said.

Clinton said, "Wherever tyrants deny the legitimate demands of their own people, we need to work together to send them a clear message: You cannot hold back the future at the point of a gun."

While promoting democracy, she warned that Europe could not ignore backsliding in its own backyard, decrying limits being placed on press and religious freedoms. She did not identify trouble spots by name but was referring to Russia, Belarus and Hungary.

"It is not credible to preach democracy elsewhere unless we also protect it within our community," she said. "The trappings of democracy are not enough."

Clinton said she had no doubt that Europe would overcome its economic woes. But she said it was imperative for a common agenda and enhanced cooperation to reinforce recoveries on both sides of the Atlantic.

"We are confident that Europe has the will and the means to cut its debt, build the necessary firewalls, create growth, and restore liquidity and market confidence," she said. But, she added: "As Europe emerges from crisis, we have to work harder to reinforce each other's recoveries. As deep as our economic relationship is, it has not yet lived up to its potential."

To boost the recovery, she said the United States and Europe need to team up to fight unfair trade practices.

Meanwhile, U.S. Defense Secretary Leon Panetta said Europe is America's security partner of choice for military operations and diplomacy around the world. He said Europe will continue to be a central U.S. defense interest, even as the administration withdraws two of the four Army brigades stationed on the continent.

In his remarks to the conference, Panetta emphasized the positive. He noted that the U.S. is building a missile defense system in Europe, including a radar installation in Turkey and missiles to be stationed in Romania and Poland. He said four U.S. ships capable of shooting down missiles will be stationed at Rota, Spain.

And he said that while two Army brigades will leave Europe, that will be partly offset by a new arrangement in which smaller Army units will rotate to the continent for temporary training assignments. This was announced in January as part of a new U.S. defense strategy.

These changes, he said, amount to a U.S. "vote of confidence" in the future of NATO.

Panetta's remarks were aimed at countering a perception that the administration's added focus on security challenges in Asia and the Middle East is weakening U.S. partnerships in Europe.

"In all, the steps Europe can expect from the United States amount to a vote of confidence from Washington in the future of the alliance, especially in a period of fiscal austerity," he said.

He noted that the U.S. military will still have a larger presence in Europe than in any other region of the world.

"That's not only because the peace and prosperity of Europe is critically important to the United States, but because Europe remains our security partner of choice for military operations and diplomacy around the world," he said.

The Obama administration has sought to limit the diplomatic fallout from its decision to pull the Army brigades out of Europe ? a move in line with an overall shrinking of the Army from 570,000 soldiers to 490,000. One brigade is scheduled to pull out of Europe in 2013 and another in 2014.

Missile defense will become a central feature of the U.S. military presence in Europe. Officials in recent days confirmed that the missile defense system's headquarters will be at Ramstein air base in Germany.

Panetta also spoke optimistically about the outlook for stability in Afghanistan, where the Taliban insurgency persists.

He said the U.S. hopes Afghan security forces will be ready to take the lead combat role throughout the country "sometime in 2013." At that point, U.S. and NATO forces will shift to a support role, he said, while remaining prepared to engage in combat if necessary.

At the conclusion of the NATO defense ministers meeting he attended in Brussels on Thursday and Friday, Panetta said there is a broad consensus among the allies that 2013 should be the year for such a transition out of a lead combat role. But France takes a different view, arguing that international forces should withdraw all combat forces from Afghanistan next year.

(AP) ? Fans of "Soul Train" boogied down Broadway wearing afro wigs and bell bottoms on Saturday while others recounted their favorite episodes at a Harlem meeting hall in tribute to the TV show's late creator, Don Cornelius.

About 100 dancers descended on Times Square in a "flash mob" organized through the Internet. As startled tourists looked on, they recreated one of the show's "Soul Train lines" in which people would take turns dancing toward a TV camera while showing off their most outrageous moves.

"Don Cornelius was a big influence in my life, and I just wanted to pay tribute," said disc jockey Jon Quick, as he held up a speaker blasting disco grooves. "He was playing the music that nobody else wanted to play. He was an amazing man."

Cornelius, 75, died of a self-inflicted gunshot wound on Wednesday. He had suffered from health problems, a difficult divorce, and had pleaded no contest to a misdemeanor spousal battery charge in 2009.

But on Saturday fans praised Cornelius' vision in creating, hosting and selling "Soul Train" to television stations that were originally skeptical about programming aimed at blacks. The show aired from 1971 to 2006.

"Don Cornelius brought soul to the whole world," said Ramona Hamm, 37, who came to Times Square with her 9-year-old daughter, Kayla Charles. The dancers bounced down Broadway for about 45 minutes until police told the party to disperse.

In Harlem, the Rev. Al Sharpton, a civil rights activist, led a tribute to Cornelius as part of the weekly community meeting at the headquarters of his National Action Network. In 1974, a 19-year-old Sharpton appeared on "Soul Train" to present an award to his mentor James Brown, the legendary soul singer.

Singer Roberta Flack said Cornelius was an inspiration to other black performers and entrepreneurs.

"He didn't have a great big light telling him, 'Go over here, don't go over there, watch where you step, there's a hole right there,'" Flack said. "He stepped."

Former "Soul Train" dancer Tyrone Proctor recalled how he hid in the trunk of a friend's car to get through the gates of the studio where "Soul Train" was filmed in 1972. Cornelius liked his dance moves and let him stay, dubbing Proctor "The Bone" because he was so skinny.

"He turned us into stars," Proctor said. Moves that "Soul Train" dancers developed spread nationwide and are now staples of music videos and pop concerts.

"Blocking, popping, ticking, waacking, punking ? when Madonna does what she does at the Super Bowl, you'll see some of these things done there," Proctor said. "Don Cornelius created all of this. It came out of his mind."

Fans recalled tuning in to see "Soul Train's" cartoon train chugging across their television screens. When Flack recalled Cornelius' stiff-necked delivery, the Harlem crowd of about 300 people laughed knowingly.

William "A.J. Dynamite" Aponte, a keyboard player, said he was ecstatic as a kid when his idol, Elton John, appeared on the show. He says the appearance showed that people of all races could find common ground in music.

"He sang 'Benny and the Jets,' and I thought it was so great because Elton John is not black, he's white and he's British," Aponte said. "It influenced me to want to do music."

Proctor said Cornelius was also generous. When Proctor won a car on "American Bandstand," ''Soul Train's" competition, Cornelius paid the $334.25 in taxes so the struggling dancer could receive his award.

"He wrote the check out and that was it, no questions asked. He just said, 'Go get the car,'" Proctor said.

Proctor and other speakers said they were shocked when they heard that Cornelius had committed suicide. Author Terrie Williams named other black performers who had killed themselves and said Cornelius' death should be a warning for victims of depression to seek help.

"One of the things that Don's death brought us to is that we've got to look in the mirror before we end up in this kind of situation," Sharpton told the crowd.
